Specialized compounds known as PROTAC (PROteolysis TArgeting Chimeras) ligands are intended to break down particular proteins in cells. These molecules are made up of two crucial parts: an E3 ubiquitin ligase recruiter and a ligand that binds to the target protein. Selectivity is ensured by carefully selecting the ligand that binds to the target protein to fit a particular pocket or binding site on the protein's surface. The other ligand attracts an E3 ubiquitin ligase, an enzyme in charge of ubiquitin-tagging proteins. Ubiquitin molecules are bound to the target protein, designating it for breakdown, when the PROTAC molecule brings the target protein and the E3 ligase close together. The cell's proteasome machinery recognizes the ubiquitinated protein and breaks it down, effectively removing it from the cellular environment. This targeted protein degradation approach shows promise in drug discovery and the development of new therapeutic strategies.
