Description | TIM-3 (T-cell immunoglobulin and mucin domain-containing protein 3), also known as HAVCR2 (hepatitis A virus cellular receptor 2), encoded by the HAVCR2 (TIM-3) gene, is a member of the TIM family of immunoregulatory proteins. The proteins of this family share a common structural organization consisting of an amino-terminal immunoglobulin variable domain with five atypical cysteines, a mucin stalk, a transmembrane domain, and a cytoplasmic tail. As a transmembrane receptor, TIM-3 has been found to have a variety of different ligands that bind to different regions on the extracellular immunoglobulin V domain of TIM3. By binding to its ligands such as Gal-9 and PtdSer, TIM-3 has been shown to modulate immune responses in autoimmunity and cancer. It has been reported that TIM-3 is mainly expressed on activated CD8+ T cells and inhibits macrophage activation after inhibition of PD-1. TIM-3 expression is upregulated in tumor infiltrating lymphocytes of different types of cancer. As an immune checkpoint, TIM-3 cooperates with the PD-1 receptor to down-regulate the activity of anti-tumor T cells, and the combination of ICI targeting TIM-3 and PD-1 has a significantly better effect on tumor prevention. This product is designed as a tool for the delivery and expression of TIM-3-TF mRNA for research. The product leverages the lipid nanoparticle (LNP) technology platform for simple and efficient delivery of TIM-3-TF mRNA to a variety of mammalian cells in vitro and in vivo. The LNPs used are formulated with SM-102, DSPC, cholesterol and DMG-PEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery. The TIM-3-TF fusion protein is approximately 32 kD, consisting of mature TIM-3 (280 amino acids) and a C-terminal TF tag (15 amino acids) |
