DescriptionExosome extraction and purification is the first step of exosome research, next, in addition to exosome identification, exosome quantification is also an important part before downstream experiments. At present, exosome quantification is mainly realized by NTA (nanoparticle tracking assay), which has more applications, i.e., through the light scattering of particles in the solution to determine the concentration of particles and the distribution of different particle sizes, but it can''t distinguish between exosomes and microvesicles and other particles, which can be stained by the plasma membrane of exosomes and microvesicles, and can be used to accurately determine the concentration of exosomes in the solution by fluorescence NTA (fNTA); in addition In addition, the concentration of exosomes can also be determined by flow cytometry, etc. These methods are complicated and costly, and require expensive instrumentation. Exosome ELISA (EXO-ELISA), i.e. Exosome Capture and Quantification Kit, adopts double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) technology, where the enzyme marker plate is pre-coated with antibodies against exosome surface antigen, which can directly capture exosome particles from samples and standards, and then the Biotin-labeled detection antibody is added, and the unbound substances are washed to remove, and then horseradish is added to the sample. After washing to remove unbound material, horseradish peroxidase-labeled Streptavidin-HRP is added, and after washing, TMB is added to the color development substrate solution, and the color is developed by avoiding light.
EXO-ELISA can capture and isolate specific total exosomes or subpopulations of exosomes in a simple and rapid manner, and the concentration of exosomes in a sample can be determined by using exosome standards; it is more convenient to use, and it can detect the expression of multiple proteins of exosomes in the same sample, or screen for changes in the expression of a particular protein in a large number of samples.
