Exosome-Mimetic Nanovesicles Production

Introduction to exosome-mimetic nanovesicles

Exosomes have been used as a promising carrier for therapeutic molecular delivery. However, if exosomes are used in biological engineering or clinic, the native production is relatively limited and does not meet the need[1]. Compared with exosomes which are passively secreted by cells, exosome-mimetic nanovesicles are artificial man-made vesicles by serial extrusions of cells through polycarbonate membranes (Figure 1 A).

The specific features of EMNs includes:

Almost 100 times of the production yield compared with natural exosomes generation[2];

  • With natural targeting ability of cells by maintaining the topology of plasma membrane proteins, the properties is similar to natural exosomes[3];
  • The drug encapsulation process can be happened at the same time with the extrusion.

The applications of EMNs includes:

  • Deliver nucleic acid drug and chemotherapeutics, et al.;
  • Affinity and specific targeting of tissues and organs;
  • Substitute exosomes with EMNs for biofunctional or clinical applications.

Besides the methods described in Figure 1 , CD Bioparticles has improved and optimized the preparation process of EMNs by membrane extraction and extrusion, which make the composition EMNs simpler without any original containing for better drug loading, no matter at a same time with the extrusion or after (Figure 2).
