DescriptionThis kit avoids the tedious gene cloning step and directly modifies the target proteins on the surface of extracted exosomes by click chemistry. First, the click chemistry linker A molecule is covalently coupled to the exosome surface to form exo-linker A, and the click chemistry linker B molecule is covalently coupled to the target protein to form pro-linkerB, and the remaining linker molecules are removed by purification of each molecule. Then exo-linker A and pro-linker B were incubated, and a fast and efficient click chemistry reaction occurred between the two linkers, which coupled the target protein to the exosome surface in the form of covalent bond, and finally the remaining pro-linker B was removed by using a SEC molecular exclusion column, which resulted in a large amount of pure modified exosomes. With high efficiency of surface modification, mild reaction conditions, the reaction can take place under physiological conditions, good reaction specificity, no harmful by-products, no need to add additional catalysts or other chemicals, no damage to the membrane structure of exosomes, and no impact on the biological activity and function of the target proteins, this kit provides a highly efficient and simple means of modification of targeted exosomes, which can realize the large-scale preparation of targeted exosomes.
