TPT, a water-soluble analog of camptothecin, specifically inhibits the activity of topoisomerase I by stabilizing the topoisomerase I-DNA complex, resulting in lethal DNA strand breaks. However, like all camptothecins, TPT undergoes pH-dependent hydrolysis of the lactone ring to form a relatively inert carboxylate salt in aqueous solution. This process acts on cells throughout the patient's body causing serious side effects. One potential solution to this problem is to encapsulate TPT in liposomes. Liposomes accumulate preferentially at tumor sites due to their ability to extravasate through "pores" or "defects" in the capillary endothelium.
